Manganese-52 is a promising positron-emitting isotope for PET: • Half-life: 5.59 days — allows tracking of slow biological processes (antibodies, nanoparticles) • Positron emission (Eavg=575 keV) — high PET resolution • Used for labeling antibodies, peptides, and cells • Important element for imaging pancreatic function and beta-cells ⚠️ CRITICAL: • Manganese-54 impurity ≤0.1% — Mn-54 (T½=312 days) is a long-lived gamma emitter. Even trace amounts create significant radiation burden to the patient over months and years. • Manganese-56 impurity ≤1.0% — Mn-56 (T½=2.6 h) has high-energy gamma lines, increasing dose to staff and patient in the first hours after administration. • Radiochemical purity ≥95% — Manganese must be stably chelated (e.g., NOTA, DOTA). Free manganese is rapidly taken up by the liver and pancreas, distorting the image. • Free manganese ≤5% — High levels of free Mn2+ lead to non-specific accumulation. Key features: • Control of long-lived Mn-54 impurity (critical for patient safety) • Control of short-lived Mn-56 impurity (affects initial dose) • Assessment of free manganese level (affects biodistribution) • Support for PET tracers and research applications Critical parameters: • Radionuclidic purity: ≥99.0% • Manganese-54 impurity: ≤0.1% • Manganese-56 impurity: ≤1.0% • Radiochemical purity: ≥95.0% • Free manganese: ≤5.0% • pH: 4.0–8.0 • Endotoxins: ≤175 EU/vial • Sterility: no microbial growth • Heavy metals: ≤10 ppm 💡 Usage tips: Radionuclidic purity is controlled by gamma spectrometry (Mn-52 peaks: 744, 935, 1434 keV; Mn-54: 834 keV; Mn-56: 847, 1811, 2113 keV) Radiochemical purity is determined by TLC or HPLC Due to the long half-life of Mn-52, control of Mn-54 impurity is especially important to minimize integral dose Manganese is an essential trace element, but toxic to the nervous system in high doses ⚠️ Note: Mn-52 is produced mainly on a cyclotron (via Cr-52(p,n)Mn-52 or NatCr(p,x)Mn-52). When using natural chromium, various manganese isotopes are formed, so purification from Mn-54 and Mn-56 is a critical production step.
