Iodine123IoflupaneQualityChecker
Iodine-123 Ioflupane
Utility description: Iodine-123 Ioflupane
Iodine-123 Ioflupane Quality Checker — Ioflupane I-123 (DaTscan®)
ℹ️ Utility checks critical parameters of the radiopharmaceutical for dopamine transporter imaging:
• Radiochemical purity (≥95%)
• Free Iodine-123 impurity (≤5%)
• Radionuclidic impurity Iodine-124 (≤0.3%) — CRITICAL!
• Radionuclidic impurity Iodine-125 (≤0.1%)
• Specific activity (≥100 GBq/μmol)
• pH, endotoxins, sterility, residual ethanol
⚠️ CRITICAL: I-124 impurity >0.3% is unacceptable! I-124 emits positrons and high-energy gamma rays, significantly increasing patient dose and degrading image quality (noise).
Free iodine accumulates in the thyroid and stomach, creating artifacts.
Usage:
Iodine123IoflupaneQualityChecker.exe → demo mode (console output)
Iodine123IoflupaneQualityChecker.exe input.csv output.json → evaluate your data
Input format:
BatchNumber,RadiochemicalPurityPercent,FreeIodine123Percent,RadionuclidicPurityI124Percent,RadionuclidicPurityI125Percent,SpecificActivityGBqPerUmol,pH,EndotoxinsEU,SterilityTest,EthanolPercent,AppearanceScore
Example (sterility: 0=sterile, 1=contaminated, appearance: 0=normal):
I123-IOF-2026-001,98.5,1.0,0.1,0.05,150.0,7.0,40.0,0,5.0,0
— WHY IS THIS NEEDED?
Ioflupane I-123 (DaTscan®) is used for diagnosing Parkinson's disease and other parkinsonian syndromes.
• I-123 (T½=13.2 hours) binds to presynaptic dopamine transporters (DAT) in the striatum.
• Allows differentiation of neurodegenerative parkinsonism (reduced uptake) from essential tremor (normal uptake).
⚠️ CRITICAL:
• Radiochemical purity ≥95% — ensures specific binding to DAT.
• Free Iodine-123 ≤5% — requires thyroid blocking with potassium iodide prior to administration if impurity is high.
• I-124 impurity ≤0.3% — I-124 (T½=4.18 days) has high positron yield, increasing dose and creating noise.
• Specific activity ≥100 GBq/μmol — necessary to prevent cold ligand from competing for receptors, which would weaken the signal.
• pH 6.0–8.0 — physiological compatibility.
• Ethanol ≤10% — solvent for the hydrophobic ligand.
Key features:
• Strict control of radionuclidic impurities (I-124/I-125) characteristic of cyclotron-produced I-123.
• Specific activity control to prevent receptor saturation.
• Appearance check (clarity/color) as the product is light-sensitive.
Critical parameters:
• Radiochemical purity: ≥95.0%
• Free Iodine-123: ≤5.0%
• I-124 impurity: ≤0.3%
• I-125 impurity: ≤0.1%
• Specific activity: ≥100 GBq/μmol
• pH: 6.0–8.0
• Endotoxins: ≤175 EU/vial
• Sterility: no microbial growth
• Ethanol: ≤10.0%
💡 Usage tips:
1. RCP is controlled by HPLC or TLC.
2. Radionuclidic purity is determined by gamma spectrometry (I-124 peaks: 602 keV, 1691 keV; I-123: 159 keV).
3. Patients must be prescribed perchlorate or potassium iodide 1 hour before injection to block the thyroid.
4. Protect the product from light (photosensitive).
⚠️ Note: Production of I-123 is often accompanied by the formation of I-124 via the (p,2n) reaction on tellurium-124 if proton energy is too high. Control of I-124 is a marker of cyclotron irradiation quality.input.csv
