Ga68DOTATATEGeneratorBasedQC
Ga-68 DOTATATE Generator Based QC
Ga-68 DOTATATE Generator Based QC — Quality control of Ga-68 DOTATATE based on generator
ℹ️ Utility checks critical parameters of Ga-68 DOTATATE:
- Radiochemical purity (≥95%)
- Free and colloidal Ga-68 (≤5% and <2%)
- Ge-68 breakthrough (<0.001%) — CRITICAL!
- Specific activity, pH, endotoxins, sterility
⚠️ CRITICAL: Ge-68 breakthrough (T½=271 days) creates long-term burden!
Ga-68 DOTATATE binds to somatostatin receptors (SSTR).
Free gallium accumulates in liver, masking metastases.

— WHY IS THIS NEEDED?
Ga-68 DOTATATE is gold standard for PET diagnosis of neuroendocrine tumors (NETs).
- Binds to somatostatin receptor subtypes (SSTR2, SSTR5).
- Ga-68 is obtained from Ge-68/Ga-68 generator (Ge-68 half-life = 271 days).
- Requires high radiochemical purity for specific tumor accumulation.
- Generator method allows daily production without cyclotron.
⚠️ CRITICAL:
- Radiochemical purity ≥95% — guarantee of specific binding to SSTR.
- Free Ga-68 ≤5% — free ions accumulate in liver and bones.
- Colloidal Ga-68 <2% — colloids are taken up by liver and spleen macrophages.
- Ge-68 breakthrough <0.001% — Ge-68 is long-lived and creates unnecessary radiation burden.
- Specific activity must be high for effective targeting.
- pH 3.5-5.5 — optimal media for Ga-DOTATATE complex stability.
- Residual solvents (ethanol from generator eluate) must be minimal (<500 ppm).
- Endotoxins <175 EU/mL — standard for parenteral drugs.
- Sterility — MANDATORY.
Key features
- Strict control of Ge-68 breakthrough characteristic of generator method.
- Separate accounting of free and colloidal gallium.
- Specific activity assessment for effective receptor binding.
- Compliance with GMP and Pharmacopoeia requirements for Ga-68 radiopharmaceuticals.
- Comprehensive safety check (endotoxins, sterility, pH, solvents).
Critical parameters
- Radiochemical purity: ≥95.0%
- Free Ga-68: ≤5.0%
- Colloidal Ga-68: <2.0%
- Specific activity: ≥10.0 GBq/mg
- Radionuclidic purity: ≥99.9%
- Ge-68 breakthrough: <0.001%
- Residual solvents: <500 ppm
- pH: 3.5–5.5
- Endotoxins: <175 EU/mL
- Sterility: no growth
💡 Usage tips:
1. Radiochemical purity determined by TLC or HPLC.
2. Ge-68 breakthrough controlled by gamma spectrometry few days after elution (when Ga-68 decays).
3. Synthesis usually performed on automatic modules or manually using kits.
4. Generator eluate contains ethanol, which must be considered in residual solvent calculation.
5. Drug must be used within few hours due to short Ga-68 half-life (68 min).
⚠️ Note: Ga-68 DOTATATE is vital tool for NET diagnosis. Generator quality and synthesis process directly affect ability to detect small metastases. Ge-68 breakthrough is unique risk of generator systems requiring constant monitoring.
