Fluorine-18 Amyloid Quality Checker — Fluorine-18 Amyloid Tracers for Alzheimer's Disease PET Imaging

ℹ️  Utility checks critical parameters of F-18 amyloid imaging agents (Florbetapir, Flutemetamol, Florbetaben):
    • Radionuclidic purity (≥99.9%)
    • Radiochemical purity (≥95%)
    • Free Fluoride-18 impurity (≤5.0%)
    • Precursor/Cold drug impurity (≤2.0%)
    • Specific activity (≥50 GBq/μmol)
    • Residual solvents (ethanol), pH, endotoxins, sterility

⚠️  CRITICAL: Free Fluoride-18 >5.0% → accumulation in skull bones, cortical artifacts!
    Low specific activity → receptor saturation, reduced plaque contrast!

— WHY IS THIS NEEDED?
F-18 agents (Florbetapir, Flutemetamol, Florbetaben) are used for Positron Emission Tomography (PET) of the brain:
• Target: Visualization of beta-amyloid neuritic plaques — a key marker of Alzheimer's Disease
• F-18 Half-life: 109.7 minutes — optimal for synthesis and delivery
• Mechanism: Binding to fibrillar amyloid in brain tissue

⚠️  CRITICAL:
• Radiochemical purity ≥95% — ensures specific binding to amyloid, not plasma proteins or other structures
• Free Fluoride ≤5.0% — [18F]fluoride is actively taken up by bone tissue. In brain scanning, this creates high background in the skull, which can be misinterpreted as cortical uptake (false positive)
• Specific activity ≥50 GBq/μmol — necessary to inject sufficient activity with minimal mass to avoid disrupting binding equilibrium
• Residual Solvents (Ethanol) — controlled per ICH Q3C, as they are used in purification (SPE)
• pH 4.5–8.5 — physiological range
• Endotoxins ≤175 EU/vial — safety standard
• Sterility — MANDATORY

💡 Usage tips:
1. Radiochemical purity and free fluoride content determined by TLC or HPLC
2. For TLC, systems such as acetonitrile/water (mobile phase) and silica gel are often used
3. Specific activity calculated from activity to molar amount ratio (HPLC-UV)
4. Due to short F-18 half-life, all measurements must be performed rapidly

⚠️ Note: Interpretation of amyloid scans requires high signal-to-noise ratio. Even small amounts of free fluoride or low specific activity can lead to diagnostic errors in assessing cortical binding.
