Sarcophagines (Sar) form clathrate complexes with copper, possessing exceptional kinetic and thermodynamic stability.

— WHY IS THIS NEEDED?
[64Cu]Cu-Sarcophagine is a class of radiopharmaceuticals where Copper-64 is encapsulated in a macrocyclic "cage" (sarcophagine):
• Exceptional Stability: The complex does not dissociate in vivo, preventing transchelation of copper by blood proteins (ceruloplasmin)
• Rapid Labeling: Complexation reaction proceeds rapidly at room temperature
• Applications: Oncology (labeled antibodies, peptides), inflammation imaging, cardiology
• Advantage: Absence of liver accumulation characteristic of free copper or less stable complexes (e.g., DOTATATE)
⚠️ CRITICAL:
• Radiochemical purity ≥98% — due to high labeling efficiency, quality standards are higher than for open chelators
• Free Copper ≤1.0% — strict limit, as even traces of free copper distort biodistribution
• Specific activity ≥30 GBq/μmol — ensures high sensitivity with low mass
• pH 4.5–8.5 — physiological range
• Endotoxins ≤175 EU/vial — safety standard
• Sterility — MANDATORY

💡 Usage tips:
1. Radiochemical purity determined by HPLC or TLC
2. TLC systems effectively separating cationic [Cu-Sar]2+ complex and free copper are often used
3. Specific activity calculated from activity to conjugate molar amount ratio
4. Complex stability allows delayed imaging (up to 24-48 hours) without signal loss due to complex decay
⚠️ Note: Sarcophagines prevent the copper ion from escaping the coordination sphere. This makes them ideal for long-term in vivo studies, where conventional chelators (DOTA, NOTA) may show some degree of demetallation.input.csv
