Copper64ATSMQualityChecker
Copper-64 ATSM
Utility description: Copper-64 ATSM
Copper-64 ATSM Quality Checker — Copper-64 ATSM for PET Hypoxia Imaging
ℹ️ Utility checks critical parameters of [64Cu]Cu-ATSM:
• Radionuclidic purity (≥99.9%)
• Radiochemical purity (≥95%)
• Free Copper-64 impurity (≤2.0%)
• ATSM Ligand impurity (≤5.0%)
• Specific activity (≥10 GBq/μmol)
• pH, endotoxins, sterility, heavy metals
⚠️ CRITICAL: Free Copper-64 >2.0% → accumulation in liver/kidneys, image degradation!
Specific activity <10 GBq/μmol → reduced hypoxia contrast!
Usage:
Copper64ATSMQualityChecker.exe → demo mode (console output)
Copper64ATSMQualityChecker.exe input.csv output.json → evaluate your data
Input format:
BatchNumber,RadionuclidicPurityPercent,RadiochemicalPurityPercent,FreeCopperPercent,ATSMLigandImpurityPercent,pH,EndotoxinsEU,SterilityTest,HeavyMetalsPPM,SpecificActivityGBqPerUmol
Example (sterility: 0=sterile, 1=contaminated):
CU64-ATSM-2026-001,99.95,98.5,0.5,1.0,7.2,50.0,0,2,25.0
— WHY IS THIS NEEDED?
[64Cu]Cu-ATSM is a lipophilic copper-64 complex used in Positron Emission Tomography (PET) for tissue hypoxia imaging:
• Cu-64 Half-life: 12.7 hours — optimal for logistics and scanning
• Mechanism: Penetrates cells, reduced under hypoxic conditions, and trapped intracellularly
• Applications: Oncology (aggressive tumor detection), Cardiology (myocardial ischemia), Neurology
⚠️ CRITICAL:
• Radiochemical purity ≥95% — ensures signal comes from ATSM complex, not free copper
• Free Copper ≤2.0% — free Cu-64 is rapidly taken up by ceruloplasmin and accumulates in the liver, creating background noise and potential toxicity
• Specific activity ≥10 GBq/μmol — necessary to prevent saturation of transport mechanisms by cold metal
• pH 4.5–8.5 — physiological range for IV administration
• Endotoxins ≤175 EU/vial — safety standard for parenterals
• Sterility — MANDATORY
Key features:
• Control of specific impurity — free Copper-64
• Assessment of specific activity as a key efficacy parameter
• Radionuclidic purity check (exclusion of long-lived satellite isotopes)
Critical parameters:
• Radionuclidic purity: ≥99.9%
• Radiochemical purity: ≥95.0%
• Free Copper-64: ≤2.0%
• ATSM Ligand: ≤5.0%
• Specific activity: ≥10.0 GBq/μmol
• pH: 4.5–8.5
• Endotoxins: ≤175 EU/vial
• Sterility: no microbial growth
• Heavy metals: ≤10 ppm
💡 Usage tips:
1. Radiochemical purity determined by TLC or HPLC with radioactive detector
2. Free copper content controlled by TLC (solvent systems: acetonitrile/water or citrate buffer)
3. Specific activity calculated from total activity and molar amount of ligand (determined by HPLC-UV)
4. Due to 12.7h half-life, all measurements must be performed promptly
⚠️ Note: Cu-ATSM is a redox-sensitive indicator. Presence of reducers or oxidizers in the preparation can distort in vivo results. Control of ligand purity and absence of foreign metals is critical for complex stability.input.csv
