Sr-89 Chloride Palliation QC — Quality control of Sr-89 Chloride for palliative therapy

ℹ️ Utility checks critical parameters of Sr-89 Chloride:

  • Radionuclidic purity (≥99.9%)
  • Sr-85 impurity (<0.01%)
  • Chemical purity and aluminum content
  • pH, osmolality, endotoxins, sterility

⚠️ CRITICAL: Sr-85 (T½=64.8 days) creates additional radiation burden!

Sr-89 Chloride is used for palliative therapy of bone pain.

Drug mimics calcium and incorporates into bone tissue.

— WHY IS THIS NEEDED?

Sr-89 Chloride (Metastron) is drug for palliative therapy of bone metastasis pain.

  • Sr-89 has 50.5-day half-life and emits high-energy beta particles.
  • Mimics calcium and selectively accumulates in areas of increased bone turnover.
  • Produced by neutron activation of enriched Sr-88 in reactor.
  • Requires high radionuclidic and chemical purity.

⚠️ CRITICAL:

  • Radionuclidic purity ≥99.9% — guarantee of absence of other strontium isotopes.
  • Sr-85 impurity <0.01% — Sr-85 has 64.8-day half-life and creates unnecessary radiation burden.
  • Chemical purity ≥99.0% — absence of other cations that may compete for bone binding.
  • Aluminum <10 ppm — toxicity and possible effect on biodistribution.
  • pH 4.0-7.5 — physiological range.
  • Osmolality 250-350 mOsm/kg — isotonicity.
  • Endotoxins <175 EU/mL — standard for parenteral drugs.
  • Sterility — MANDATORY.

Key features

  • Strict control of Sr-85 impurity characteristic of reactor production.
  • Assessment of chemical purity and aluminum content.
  • Osmolality and pH check for administration safety.
  • Compliance with GMP and Pharmacopoeia requirements for Sr-89 radiopharmaceuticals.
  • Comprehensive safety check (endotoxins, sterility, pH, osmolality).

Critical parameters

  • Radionuclidic purity: ≥99.9%
  • Sr-85 impurity: <0.01%
  • Other radionuclides: <0.01%
  • Chemical purity: ≥99.0%
  • Aluminum: <10 ppm
  • pH: 4.0–7.5
  • Osmolality: 250–350 mOsm/kg
  • Endotoxins: <175 EU/mL
  • Sterility: no growth

💡 Usage tips:

1. Radionuclidic purity determined by gamma spectrometry.

2. Chemical purity and aluminum content controlled by atomic spectroscopy methods.

3. Drug is stable for long time due to simple chloride chemical form.

4. Sr-89 does not emit gamma quanta, which makes imaging difficult but reduces radiation burden on staff.

5. Therapeutic effect usually occurs 1-3 weeks after administration.

⚠️ Note: Sr-89 Chloride is one of oldest radiopharmaceuticals for palliative therapy. Its simplicity (inorganic salt) requires special attention to radionuclidic purity, as any impurities of other strontium isotopes will also accumulate in patient's bones for many years.
