— WHY IS THIS NEEDED?
Ulipristal acetate — selective progesterone receptor modulator (SPRM):
• Emergency contraception (Ella, EllaOne — up to 120 hours after unprotected intercourse)
• Treatment of uterine fibroid symptoms (Esmya — with EMA restrictions due to liver injury risk)
⚠️ CRITICAL:
• Specific rotation +105.0° to +115.0° — key identification parameter (in methanol)
• Melting point 180.0–185.0°C (with decomposition) — confirms structure and purity
• UV ratio A240/A280: 1.8–2.2 — confirms structure (maximum at 240 nm)
• Ulipristal ≤0.5% — HYDROLYSIS PRODUCT OF ACETATE GROUP! Forms on moisture contact, reduces activity and alters pharmacokinetics
• Epimer ≤0.5% — STEREOISOMER! Affects receptor binding and safety
• Total impurities ≤1.0% — critical due to narrow therapeutic window and liver injury risk
⚠️ Special attention: Ulipristal acetate is EXTREMELY sensitive to moisture and hydrolysis! Requires storage in airtight, moisture-protected packaging at temperature not exceeding 25°C. Analysis must be performed under humidity-controlled conditions! Ulipristal impurity (free alcohol) is especially critical — even minor amounts indicate degradation and disrupt action profile. Loss on drying control (≤0.5%) is mandatory. In 2020, EMA suspended ulipristal acetate for fibroid treatment due to serious liver injury risk — substance quality is critical for risk minimization!

1. Scope

The Ulipristal Acetate Quality Checker utility is used for Ulipristal Acetate. The actual input.csv is the source of truth for the input structure; control criteria are defined by the executable and the domain description.

The utility does not use machine learning; decisions are produced by deterministic rules.

9. OQ/PQ test scenarios

10. QA/QC, CSV and change control

  • Before production use, the client records executable version, checksum, specification/monograph version, test CSV, expected JSON and IQ/OQ results.
  • Column names must not be changed without updating the validator and test scenarios.
  • The source CSV, output.json and utility version should be stored together as an evidence package.
  • Any change in control rules must go through change control and repeated OQ scenario verification.
