Utility description: Topical Formulation Spot-On

Topical Formulation Spot-On Quality Checker — Control of Spot-On Topical Forms

ℹ️ Utility assesses the quality of "spot-on" dosage forms for pets according to internal standards of manufacturers (Zoetis, Elanco, Merck) and general GMP requirements:
     • Fill Weight Accuracy: Deviation no more than ±5% from nominal.
     • Active Substance Content (Assay): 90-110% of declared.
     • Viscosity: Critical parameter for ensuring flow through the narrow pipette tip and subsequent spreading on the animal's skin.
     • Evaporation Loss: Control of solvent volatility. High evaporation leads to drug crystallization on fur (skin irritation) and dose loss.
     • Packaging Integrity: Check of blister/pipette seal.

⚠️ CRITICAL:
     • Crystallization: If the solvent evaporates too quickly, the active ingredient (e.g., fipronil or selamectin) may crystallize on the fur instead of distributing into the skin lipid layer.
     • Dosing: For cats and puppies, volumes are very small (0.5–1.0 mL). An error of 0.1 mL constitutes 10-20% of the dose, which is unacceptable.

Usage:
TopicalFormulationSpotOnQualityChecker.exe → demo mode (console output)
  TopicalFormulationSpotOnQualityChecker.exe input.csv output.json → evaluate your data

Input format:
BatchNumber,ProductName,FillWeight_g,TargetWeight_g,Assay_Percent,Viscosity_cP,EvaporationLoss_Percent,FlashPoint_C,PackagingIntegrity

Example:
  SO-2026-FIP-001,Fipronil Cat,0.51,0.50,98.5,15.0,1.2,45.0,Pass

— WHY IS THIS NEEDED?
Spot-on forms are one of the most popular methods for delivering ectoparasiticides.
Quality depends not only on chemistry but also on fluid physics and packaging accuracy.
Incorrect viscosity will result in the owner being unable to squeeze out the product or it running off in a drop without spreading.

⚠️ CRITICAL:
• Viscosity: Must be optimized for storage temperature. Too thick product in winter is difficult to apply.
• Solvents: Alcohols, ethers, or glycols are often used. Their ratio affects the rate of penetration through the skin barrier.
• Packaging: Pipettes must be child-and-pet resistant but easy for the owner to open.
• Compatibility: The product must not degrade the pipette plastic or label adhesive during long-term storage.

Key features:
• Comprehensive check of physical and chemical parameters
• Solvent stability control (evaporation)
• Assessment of small volume dosing accuracy
• Generation of reports for filling production lines and LIMS LabWare
• Compliance with GMP requirements for veterinary drugs

Critical parameters:
• Fill Weight Deviation: ±5%
• Assay: 90-110%
• Viscosity: 10-50 cP (typical)
• Evaporation: < 2% per 24 hours
• Integrity: Pass

💡 Usage tips:
1. Temperature Control: Measure viscosity at 20°C and 40°C to guarantee applicability in different climatic zones.
2. Spreadability Test: Additionally assess the diameter of the drug spot on artificial skin or glass.
3. Pipette Weight: Weigh empty and filled pipettes for accurate net weight calculation.
4. Volatility: Use closed filling systems to minimize solvent loss and operator exposure.
5. Validation: The filling process must be validated for absence of leaks and dosing accuracy.

⚠️ Note: Unlike injections, where the dose enters the systemic circulation immediately, Spot-on products must overcome the skin barrier and distribute via sebaceous glands. The physicochemical properties of the vehicle are as important as the active ingredient itself.

QA/QC and change control

  • Do not rename columns without updating validator, documentation and test set.
  • Retain input.csv, output.json, executable version and checksum.
  • Before production use, perform IQ/OQ/PQ or equivalent CSV/CSA verification.
  • Critical limits shall be verified against the approved specification, registration dossier and local SOPs.
  • The utility provides structured QC decision support; final release decision remains with QA/QP and approved procedures.
