Potency Assay Animal Model Compliance Quality Checker — Control of In Vivo Potency Tests

ℹ️  Utility assesses results of biological potency tests for vaccines requiring laboratory animals (according to specific Ph. Eur., USP monographs):
     • Potency: Calculated value in International Units (IU).
     • Statistical Significance: Lower Confidence Limit (P=0.95) must exceed minimum required level.
     • Ethical Compliance: Verification of humane animal treatment principles (3R).
     • Model Validity: Control group must show expected level of disease/mortality upon challenge.

⚠️  CRITICAL: 
     • Even if mean potency is high, if Lower Confidence Limit is below limit — batch is rejected (insufficient statistical confidence).
     • Any ethical violation (Ethical Compliance Fail) invalidates the test and requires internal investigation.
     • If control group survives (low mortality in challenge), the infection model failed, and test is invalid.

— WHY IS THIS NEEDED?
For some vaccines (whole-cell pertussis, anthrax, some rabies forms), fully validated alternative in vitro methods are not yet developed.
Animal testing remains the only way to prove protective efficacy before batch release.
This is the most expensive, time-consuming, and ethically complex quality control step.

⚠️  CRITICAL:
• Statistics: Parallel-line assay or probit analysis is used to calculate ED50/LD50.
• Challenge Control: In protection assays, unvaccinated animals must die/get sick. If they survive, the vaccine is not to blame — the test is bad.
• Animal Welfare: Monitoring of pain, suffering, and distress is mandatory. Animals with severe symptoms must be euthanized per protocol.
• Alternatives (3R): Industry is actively moving to replace these tests with serological methods (in vitro potency) where possible.

💡 Usage tips:
1. Study Design: Number of animals must be sufficient for statistical power but minimal according to Reduction principle.
2. Randomization: Animals must be assigned to groups randomly to exclude systematic errors.
3. Blinding: Result assessment (e.g., symptom presence) should be done "blind" to avoid operator bias.
4. Documentation: All cases of animal distress must be documented in the observation log.
5. Trends: Track potency variability from batch to batch. High variability may indicate manufacturing issues.

⚠️ Note: This test is a production "bottleneck". It takes weeks (incubation + observation) and requires expensive vivarium facilities. Automation of data analysis helps reduce decision-making time.
