ℹ️  Utility checks critical oxidation parameters of Polysorbate 80:
   • Peroxide Value: ≤5.0 meq/kg
   • Peroxide Content: ≤100 ppm
   • Acid Value: ≤2.0 mg KOH/g
   • Color (Absorbance @ 400nm): ≤0.3 (indicator of degradation products)

⚠️  CRITICAL: High peroxides → oxidation of methionine and tryptophan in therapeutic proteins!
   This leads to loss of drug activity and increased immunogenicity.

— WHY IS THIS NEEDED?
Polysorbate 80 (Tween 80) is the most common stabilizer for protein drugs.
• Contains oleic acid (unsaturated), which easily auto-oxidizes with air oxygen.
• Auto-oxidation products (peroxides, aldehydes, acids) are toxic to proteins.
• Protein oxidation (especially Met, Trp, His) is a major cause of biopharmaceutical batch failure.
• Pharmacopoeias (Ph. Eur., USP) and biotech internal standards strictly limit peroxides.

⚠️  CRITICAL:
• Peroxide Value ≤5.0 meq/kg — standard limit for pharma grade. For sensitive proteins, <1.0 meq/kg is required.
• Acid Value ≤2.0 — increase indicates hydrolysis of ester bonds and accumulation of free fatty acids.
• Color: Fresh PS80 is pale yellow. Darkening indicates deep oxidation.
• Peroxides can accumulate during storage, especially with exposure to light and heat.

💡 Usage tips:
1. Use ferricyanide or iodometric method for peroxide determination.
2. For biopharma, enzymatic methods or HPLC with fluorescence detection are preferred (high sensitivity).
3. Store PS80 under inert atmosphere (nitrogen), in dark, and at low temperature.
4. Before use in sensitive processes, PS80 can be purified from peroxides (e.g., passing through a reducer column).
5. Regularly test each new batch of PS80 before introduction into production.

⚠️ Note: Polysorbate 80 is a complex mixture, and its quality varies significantly between manufacturers. "Bio-grade" (Polysorbate 80 NF/EP Grade for Biopharma) usually has lower levels of peroxides and impurities than standard food or cosmetic grades.

URS & FS — User Requirements and Functional Specification

This document defines the controlled interface, validation behaviour and expected output of Polysorbate80PeroxideChecker. The document is intended to reside inside the utility folder together with input.csv and the description file. Expected executable: Polysorbate80PeroxideChecker.exe.

Purpose: the utility performs deterministic verification of QC input data against approved limits, generates machine-readable output.json, and supports QC/QA review, LIMS/ELN/MES integration and CSV/CSA preparation.

QA/QC, CSV/CSA and change control
