Polyethylene Glycol Impurity Checker — Control of Polyethylene Glycol (PEG) Impurities

ℹ️  Utility checks critical safety parameters of PEG:

— WHY IS THIS NEEDED?
Polyethylene Glycols (PEG) are widely used as ointment bases, suppository bases, and tablet fillers.
• PEG synthesis involves ethylene oxide polymerization, leaving monomers (ethylene glycol) and oligomers (diethylene glycol).
• Ethylene glycol and diethylene glycol are highly toxic to kidneys and nervous system.
• Pharmacopoeias (Ph. Eur., USP) require strict control of these impurities by GC.
• Acid value indicates degree of PEG oxidation during storage (formation of aldehydes and acids).

⚠️  CRITICAL:
• Ethylene Glycol ≤10 ppm — strict limit due to high toxicity.
• Diethylene Glycol ≤2.0% (or lower for injectables) — control of reaction completeness.
• Acid Value ≤0.2 — ensures freshness and absence of oxidation products.
• PEGs are hygroscopic, so moisture affects dosage form stability.

💡 Usage tips:
1. Use GC with Flame Ionization Detector (FID) for ethylene glycol and DEG determination.
2. Derivatization or high-resolution capillary column may be needed for ethylene glycol.
3. Acid value is determined by titration. Increase indicates improper storage (air/light exposure).
4. Low molecular weight PEGs (liquids) are more prone to oxidation than solid PEGs.
5. If ethylene glycol >10 ppm is detected, batch is rejected immediately.

⚠️ Note: PEGs do not have a single molecular weight but are polymer mixtures. Therefore, control of lower glycol impurities is the main marker of polymer purification quality.

URS & FS — User Requirements and Functional Specification

This document defines the controlled interface, validation behaviour and expected output of PolyethyleneGlycolImpurityChecker. The document is intended to reside inside the utility folder together with input.csv and the description file. Expected executable: PolyethyleneGlycolImpurityChecker.exe.

Purpose: the utility performs deterministic verification of QC input data against approved limits, generates machine-readable output.json, and supports QC/QA review, LIMS/ELN/MES integration and CSV/CSA preparation.

QA/QC, CSV/CSA and change control

  • Do not rename columns without updating the validator, test set and documentation.
  • Retain input.csv, output.json, executable version, checksum and a copy of the documentation.
  • Before production use, perform IQ/OQ/PQ or equivalent risk-based CSV/CSA verification.
  • Critical limits shall be approved by the process owner and verified against the registration dossier, pharmacopoeia and local SOPs.
