Levonorgestrel Emergency Quality Checker — Emergency Contraceptive Tablets (Postinor, Escapelle) (Ph. Eur., ICH Q6A)

ℹ️ The utility checks levonorgestrel content (% of declared), impurities (3-keto and epimer), dissolution (≥80% at 30 min), tablet physical properties, microbiology (including pathogens).
ℹ️ Does not use machine learning — rule-based logic only.
⚠️ CRITICAL for emergency contraception! Dissolution ≥80% at 30 min ensures rapid absorption and efficacy within 72 hours. Strict impurity control is mandatory!

Usage:
LevonorgestrelEmergencyQualityChecker.exe → demo mode (console output)
LevonorgestrelEmergencyQualityChecker.exe input.csv output.json → evaluate your data

Input format:
BatchNumber,LevonorgestrelContent,Impurity3Keto,ImpurityEpimer,TotalImpurities,Dissolution,WeightVariationRSD,DisintegrationTime,Friability,HeavyMetals,TAMC,TYMC,EColiDetected,SalmonellaDetected,EthanolPPM,AcetonePPM

Example (EColiDetected/SalmonellaDetected: 0=not detected, 1=detected):

— WHY IS THIS NEEDED?
Levonorgestrel emergency contraceptive tablets — critically important preparation for preventing unwanted pregnancy:
• Used within 72 hours after unprotected intercourse (Postinor — 2 tablets of 0.75 mg, Escapelle — 1 tablet of 1.5 mg)
• Efficacy directly depends on absorption speed and bioavailability
• Must be available OTC — quality must be impeccable!
⚠️ CRITICAL:
• Dissolution ≥80% at 30 minutes — ensures rapid absorption and maximum efficacy in critical time window
• Levonorgestrel content 90.0–110.0% of declared — guarantees therapeutic dose
• 3-Keto impurity ≤0.5% — oxidation product, reduces activity
• Epimer (13β/17α) ≤0.5% — stereoisomer, affects pharmacokinetics
• Absence of pathogens (E. coli, Salmonella) — mandatory for oral preparations, especially during potential stress use
⚠️ Special attention: Emergency contraception is a trust medication! Even minor deviation in dissolution or active substance content can lead to inefficacy and unwanted pregnancy. Dissolution tested strictly by HPLC method (not UV!) for accurate release determination.
Critical parameters:
• Levonorgestrel content: 90.0–110.0% of declared
• 3-Keto impurity: ≤0.5%
• Epimer (13β/17α): ≤0.5%
• Total impurities: ≤1.0%
• Dissolution: ≥80.0% (at 30 min, not exceeding 100%)
• Weight variation (RSD): ≤5.0%
• Disintegration: ≤30 minutes
• Friability: ≤1.0% (for uncoated tablets)
• Heavy metals: ≤20 ppm
• TAMC: ≤1000 CFU/g
• TYMC: ≤100 CFU/g
• Absence of E. coli and Salmonella
• Residual solvents: ethanol ≤5000 ppm, acetone ≤5000 ppm
⚠️ Note: For emergency contraception, speed of action is critical! Dissolution must be rapid and complete. Tablets often have special formulation (fast-dissolving) or coating to accelerate absorption. Quality control must include dissolution testing under conditions simulating GI tract (pH 1.2, 45 rpm).
