⚠️  IMPORTANT: Ribavirin is a synthetic guanosine nucleoside analogue.
    Instead of a purine base, the molecule contains 1,2,4-triazole-3-carboxamide
    linked to β-D-ribofuranose via N-glycosidic bond.
    The triazole ring mimics the imidazole portion of purine, while the carboxamide
    group (-CONH₂) replaces guanine's amino group.
    Multiple ribose hydroxyl groups form an extensive hydrogen bonding network,
    creating a very broad absorption band at 3200–3500 cm⁻¹.

— WHY IS THIS NEEDED?
Ribavirin is a broad-spectrum antiviral agent.
• Used in combination with interferon or sofosbuvir for hepatitis C treatment.
• Mechanism: viral RNA polymerase inhibition and intracellular GTP pool depletion.
• IR spectroscopy is the primary identity confirmation method per Ph. Eur. and Russian Pharmacopoeia.
• Reference spectrum established in Ph. Eur. Ribavirin and Russian Pharmacopoeia FS.2.4.00270.18.
• InfraLUM FT-08/FT-10 instruments are widely used in Russian QC laboratories.

⚠️  CRITICAL:
• Correlation ≥0.995 — strict matching criterion with reference.
• Peak 1680 cm⁻¹ (amide C=O) — primary carboxamide group marker.
  Distinguishes ribavirin from natural nucleosides (guanosine has no amide).
  Shift to ~1650 cm⁻¹ may indicate amide hydrolysis to acid.
• Peak 1620 cm⁻¹ (triazole C=N + NH₂ bend) — heterocyclic base marker.
  Absence indicates triazole ring destruction.
• Peak 1210 cm⁻¹ (ribose C-O-C) — specific furanose ring marker.
  Distinguishes ribavirin from deoxyribonucleosides (different sugar conformation).
• Peak 1050 cm⁻¹ (C-O CH₂OH) — confirms primary alcohol group presence.

💡 Usage tips:
1. KBr tablets must be DRY — pre-dry at 120°C before use.
   Ribavirin is VERY hygroscopic! Moisture radically distorts the 3200–3500 cm⁻¹ region.
2. Sample concentration in KBr: 0.5–1.0% (w/w).
3. Record spectrum in 4000–400 cm⁻¹ range, 4 cm⁻¹ resolution, 32 scans.
4. Record background spectrum of pure KBr before each measurement.
5. For ATR method, ensure tight contact; ribavirin compresses well.
6. Reference spectrum should be obtained on the same instrument under identical conditions.
7. Prepare sample QUICKLY under low humidity conditions (<40% RH).

⚠️ Note: Ribavirin exists in two anomeric forms (α and β), but only the β-D-ribofuranose
    form is pharmacopoeial. The α-anomer shows differences in the ribose C-O-C region
    (1000–1250 cm⁻¹) and may reduce correlation.
    If α-anomer presence is suspected, perform specific rotation test or chiral HPLC.
    The carboxamide group (-CONH₂) also gives a characteristic Amide II band
    (N-H bend + C-N stretch) at ~1620 cm⁻¹, which overlaps with triazole peaks.
    This overlap is normal for ribavirin and accounted for in the reference spectrum.
    Ribofuranosyl acetate impurity (protected synthesis precursor) manifests as
    additional ester C=O peaks at ~1740 cm⁻¹. Presence of a peak in this region
    requires quantitative impurity control by HPLC.

URS & FS — User Requirements and Functional Specification

This document defines the controlled interface, validation behaviour and expected output of InfraLUM_ID_Ribavirin. The document is intended to reside inside the utility folder together with input.csv and the description file. Expected executable: InfraLUM_ID_Ribavirin.exe.

Purpose: the utility performs deterministic verification of QC input data against approved limits, generates machine-readable output.json, and supports QC/QA review, LIMS/ELN/MES integration and CSV/CSA preparation.

QA/QC, CSV/CSA and change control

  • Do not rename columns without updating the validator, test set and documentation.
  • Retain input.csv, output.json, executable version, checksum and a copy of the documentation.
  • Before production use, perform IQ/OQ/PQ or equivalent risk-based CSV/CSA verification.
  • Critical limits shall be approved by the process owner and verified against the registration dossier, pharmacopoeia and local SOPs.
