Utility description: InfraLUM ID Omeprazole

InfraLUM ID Omeprazole — Omeprazole Identification by IR Spectroscopy

ℹ️ Utility checks IR spectrum compliance with reference (Ph. Eur. / Russian Pharmacopoeia):
    • Correlation coefficient with reference spectrum: ≥0.995
    • Peak 3450 cm⁻¹ (N-H benzimidazole stretching): ±20 cm⁻¹
    • Peak 1620 cm⁻¹ (C=N/C=C aromatic): ±10 cm⁻¹
    • Peak 1270 cm⁻¹ (S=O sulfoxide): ±10 cm⁻¹ — CRITICAL MARKER
    • Peak 1210 cm⁻¹ (C-O-C ether bond): ±10 cm⁻¹
    • Peak 1050 cm⁻¹ (S-O-C / pyridine ring): ±10 cm⁻¹
    • Baseline noise: ≤0.005 Abs

⚠️ IMPORTANT: Omeprazole is a proton pump inhibitor (H⁺/K⁺-ATPase).
    Molecule contains three key fragments:
    1. Benzimidazole ring — enzyme binding site.
    2. Pyridine ring with methoxy groups — provides lipophilicity.
    3. Chiral sulfoxide bridge (-S(=O)-CH₂-) — PRODRUG.
In acidic gastric environment, sulfoxide rearranges to active
sulfenamide that covalently binds to ATPase SH-groups.
Oxidation of sulfoxide to sulfone (-SO₂-) leads to loss of activity!

Usage:
InfraLUM_ID_Omeprazole.exe → demo mode (console output)
  InfraLUM_ID_Omeprazole.exe input.csv output.json → evaluate your data

Input format:
BatchNumber,CorrelationCoefficient,Peak3450_cm1,Peak1620_cm1,Peak1270_cm1,Peak1210_cm1,Peak1050_cm1,BaselineNoiseAbs,PreparationMethod

Example (PreparationMethod: KBr / ATR / Nujol):
  OMP-ID-2026-033,0.9993,3448.0,1618.0,1272.0,1208.0,1052.0,0.0015,KBr

— WHY IS THIS NEEDED?
Omeprazole is one of the most prescribed gastroenterological drugs.
• Mechanism of action: irreversible inhibition of parietal cell H⁺/K⁺-ATPase.
• Prodrug: activated only in acidic environment (pH < 4).
• IR spectroscopy is the primary identity confirmation method per Ph. Eur. and Russian Pharmacopoeia.
• Reference spectrum established in Ph. Eur. Omeprazole and Russian Pharmacopoeia FS.2.4.00243.18.
• InfraLUM FT-08/FT-10 instruments are widely used in Russian QC laboratories.

⚠️ CRITICAL:
• Correlation ≥0.995 — strict matching criterion with reference.
• Peak 1270 cm⁻¹ (S=O sulfoxide) — PRIMARY functional group marker.
Shift to ~1320 cm⁻¹ indicates oxidation to sulfone (degradation!).
Reduced intensity may indicate reduction to sulfide.
• Peak 3450 cm⁻¹ (N-H benzimidazole) — heterocyclic core marker.
Sensitive to hydrogen bonding and moisture.
• Peak 1210 cm⁻¹ (C-O-C) — confirms ether bridge between rings.
• Peak 1050 cm⁻¹ (S-O-C/pyridine) — combined sulfoxide-methylene vibration.
• Noise >0.005 Abs renders spectrum unsuitable for identification.

Key features:
• Uses NAMED TUPLE (record struct EvalResult) —
  eliminates positional field access errors.
• Specification based on Ph. Eur. Omeprazole and Russian Pharmacopoeia FS.2.4.00243.18.
• Control of five characteristic peaks + correlation + spectrum quality.
• Sulfoxide degradation monitoring via S=O peak position.
• Full LabWare LIMS compatibility.

Critical parameters:
• Correlation Coefficient: ≥0.995
• Peak 3450 cm⁻¹ (N-H benzimidazole): 3450 ± 20 cm⁻¹
• Peak 1620 cm⁻¹ (C=N/C=C aromatic): 1620 ± 10 cm⁻¹
• Peak 1270 cm⁻¹ (S=O sulfoxide): 1270 ± 10 cm⁻¹
• Peak 1210 cm⁻¹ (C-O-C ether): 1210 ± 10 cm⁻¹
• Peak 1050 cm⁻¹ (S-O-C/pyridine): 1050 ± 10 cm⁻¹
• Baseline Noise: ≤0.005 Abs

💡 Usage tips:
1. KBr tablets must be dry — pre-dry at 120°C before use.
Omeprazole is sensitive to moisture and acid!
2. Sample concentration in KBr: 0.5–1.0% (w/w).
3. Record spectrum in 4000–400 cm⁻¹ range, 4 cm⁻¹ resolution, 32 scans.
4. Record background spectrum of pure KBr before each measurement.
5. For ATR method, ensure tight crystal-sample contact.
6. Reference spectrum should be obtained on the same instrument under identical conditions.
7. Omeprazole is photosensitive and thermolabile — prepare sample quickly at room temperature.

⚠️ Note: Omeprazole is a racemate. IR spectroscopy does NOT distinguish
R/S enantiomers. Esomeprazole (S-isomer) has an identical IR spectrum.
Use polarimetry or chiral HPLC to differentiate omeprazole from esomeprazole.
ACID DEGRADATION: upon contact with acid (even trace HCl in KBr),
omeprazole rapidly converts to sulfenamide and then to disulfide dimers.
This manifests as new peaks in the 1650–1700 cm⁻¹ region and disappearance
of the S=O peak at 1270 cm⁻¹. If this pattern is observed, the issue is
in sample preparation, not the substance. Use neutral KBr and minimize contact time.
Methoxy groups on both rings give characteristic C-H peaks at ~2840 cm⁻¹
and C-O at ~1020 cm⁻¹, serving as additional confirmatory markers.
