Fluorogenic substrates (non-fluorescent before reaction) allow high-sensitivity and selective determination of enzyme activity (proteases, lipases, esterases, phosphatases, etc.). • Lumex Fluorat instruments provide low detection limits, critical for kinetic measurements at early reaction stages. • The method is widely used in quality control of enzyme preparations, diagnostic kits, and biotechnological processes. • Control of reaction rate (kinetics) ensures measurement is performed in the initial linear stage, where rate is proportional to enzyme concentration. ⚠️ CRITICAL: • Activity must match specification (usually ±10%). • Signal-to-Noise Ratio (SNR) must be high (>10) to exclude influence of background fluorescence from solvent or glassware. • Reaction temperature is strictly controlled, as enzyme activity is highly temperature-dependent (Q10). • Inner filter effect must be excluded at high product concentrations.
