CagrilintideSemaglutide_ObesityCombinationPipelineWatcher
Cagrilintide/Semaglutide Obesity Combination Pipeline Watcher
ℹ️ Utility checks critical parameters of amylin and GLP-1 analog combination:
• Cagrilintide Assay: 90.0–110.0%
• Semaglutide Assay: 90.0–110.0%
• Impurities of each component: ≤1.0%
• Total Impurities: ≤2.0%
• Osmolality: 250–350 mOsm/kg
• pH: 7.0–8.5
• Subvisible Particles: ≤6000 per mL
• Endotoxins: ≤0.5 EU/mg
⚠️ CRITICAL: Peptide products are prone to aggregation. Particle and osmolality control is important.
Violation of component ratio may change safety profile (nausea).

— WHY IS THIS NEEDED?
Cagrilintide (amylin receptor agonist) and semaglutide (GLP-1 agonist) combination is an innovative obesity therapy:
• Cagrilintide — modified amylin analog, reduces appetite and delays gastric emptying
• Semaglutide — glucagon-like peptide-1 analog, enhances insulin secretion and reduces appetite
• Synergistic effect provides superior weight loss compared to monotherapy
• Both components are peptides requiring strict control of aggregation and degradation
⚠️ CRITICAL:
• Assay 90.0–110.0% for each component — ensures therapeutic effect
• Impurities ≤1.0% for each component — degradation products (deamidation, oxidation)
• Total Impurities ≤2.0% — overall purity indicator
• Osmolality 250–350 mOsm/kg — isotonicity for subcutaneous injection
• pH 7.0–8.5 — peptide stability and injection comfort
• Subvisible Particles ≤6000 per mL — protein aggregation control
• Endotoxins ≤0.5 EU/mg — injectable product safety
Key features:
• Simultaneous check of two peptide active substances
• Individual impurity control considering peptide degradation specifics
• Osmolality and pH control as stability and tolerability factors
• Subvisible particle (aggregate) monitoring
Critical parameters:
• Cagrilintide Assay: 90.0–110.0%
• Semaglutide Assay: 90.0–110.0%
• Cagrilintide Impurities: ≤1.0%
• Semaglutide Impurities: ≤1.0%
• Total Impurities: ≤2.0%
• Osmolality: 250–350 mOsm/kg
• pH: 7.0–8.5
• Subvisible Particles: ≤6000 count/mL
• Endotoxins: ≤0.5 EU/mg
💡 Usage tips:
1. Assay determined by HPLC with UV or fluorescence detection
2. Impurities controlled by specific HPLC methods (gradient elution)
3. Osmolality determined by cryoscopic method
4. Particles controlled by light obscuration (LO) method
5. Store at 2-8°C, do not freeze
6. If high levels of aggregates detected, check storage and transportation conditions
⚠️ Note: Combination of two peptides increases risk of physicochemical interactions, such as co-aggregation or complex formation. Cagrilintide is an acylated peptide, making it more lipophilic but also susceptible to amide bond hydrolysis. Semaglutide contains a fatty acid, which also affects its behavior in solution. Strict quality control is necessary to ensure predictable pharmacokinetics.

QA/QC and change control
- Do not rename columns without updating validator, documentation and test set.
- Retain input.csv, output.json, executable version and checksum.
- Before production use, perform IQ/OQ/PQ or equivalent CSV/CSA verification.
- Critical limits shall be verified against the approved specification, registration dossier and local SOPs.
- For combination products, document whether the specialized FDC utility or fallback component checks were used.
- The utility provides structured QC decision support; final release decision remains with QA/QP and approved procedures.
