AyurFinishedProductDissolutionChecker
Ayur Finished Product Dissolution
ℹ️ Utility evaluates release profile of marker compounds from finished Ayurvedic dosage forms with adaptive criteria for specific matrices:
• Disintegration: adaptive limits by dosage form
• Dissolution profile: 3 timepoints (30 / 60 / 120 minutes)
• Profile monotonicity: detection of matrix anomalies (swelling, gelation)
• Adaptive release limits for each form category
⚠️ WHY STANDARD PH.EUR./USP <711> TEST IS NOT APPLICABLE:
Ayurvedic dosage forms are fundamentally different from synthetic tablets:
• Guggulu Vati: contain 30-50% Commiphora mukul resin (lipophilic matrix)
→ Disintegration 45-90 min (normal!), not 15-30 min like synthetics
→ Release extended over 2-4 hours
• Vati with honey/ghee: hydrophobic binders slow water penetration
→ Extended timepoints required
• Churna (powders): absence of binder matrix
→ Rapid release, strict limits at early timepoints
Applying standard USP limits (Q=75% at 45 min) would result in
false rejection of ALL legitimate Guggulu preparations.
Usage:
AyurFinishedProductDissolutionChecker.exe → demo mode
AyurFinishedProductDissolutionChecker.exe input.csv output.json → evaluate data
Input format:
Batch,Product,DosageForm,Marker,Diss30%,Diss60%,Diss120%,Disintegration_min,Medium
Example:
AYUR-001,Yograj_Guggulu,Guggulu_Vati,Guggulsterone,35.0,62.0,85.0,45.0,SGF_then_SIF
Adaptive limits by dosage form:
• Guggulu_Vati / Resin tablets:
Disintegration ≤90 min | @30min ≥15% | @60min ≥40% | @120min ≥70%
• Vati / Tablet (honey/ghee/gum):
Disintegration ≤60 min | @30min ≥25% | @60min ≥55% | @120min ≥80%
• Capsule:
Disintegration ≤30 min | @30min ≥40% | @60min ≥70% | @120min ≥85%
• Churna / Powder:
Disintegration ≤15 min | @30min ≥50% | @60min ≥75% | @120min ≥90%
— PARAMETERS AND THEIR SIGNIFICANCE:
• Disintegration Time:
Time for complete tablet/capsule breakdown in medium.
Up to 90 min allowed for Guggulu forms due to resin matrix.
Exceedance = compression issue or excess binder.
• Dissolution @ 30/60/120 min:
Cumulative % marker release at each timepoint.
Limits adapted to specific form kinetics.
• Release Profile Consistency (monotonicity):
Profile must be monotonically increasing: D30 ≤ D60 ≤ D120.
Non-monotonicity (D60 < D30) indicates matrix anomaly:
- Resin swelling followed by collapse
- Marker precipitation upon pH change
- Gum gelation blocking diffusion
This is a unique parameter absent from standard tests.
💡 Usage tips:
1. Use biphasic medium for Guggulu: SGF (pH 1.2, 2h) → SIF (pH 6.8)
2. For lipophilic markers (guggulsterones), add 0.5-1% SLS to medium
3. Method: USP Apparatus 2 (Paddle), 50-75 rpm (reduced speed for resins)
4. Sample with medium replacement, filter through 0.45 μm
5. Establish individual limits for each new product based on 3+ validation batches
6. Correlate with clinical data: if in vivo effect exists at low in vitro release — revise limits
7. Track trends: profile drift between batches = raw material quality change (resin, honey)
⚠️ Note: This utility is the FIRST bioavailability control tool for finished
Ayurvedic dosage forms adapted to their unique matrix nature. It closes the last
gap in the QC chain: from raw material identification → safety → stability →
formula accuracy → ACTIVE COMPOUND RELEASE. Without this test, it is impossible
to guarantee that patient receives therapeutic dose even if all previous tests passed.
