AyurFermentedPreparationAlcoholAndMicrobiomeChecker
Ayur Fermented Preparation Alcohol And Microbiome
ℹ️ Utility controls quality of live fermented Ayurvedic preparations according to AYUSH standards and API Part-I:
• Self-generated ethanol: 5–12% v/v (natural preservative and extractant)
• pH: 3.0–4.5 (acidic environment suppresses pathogens, supports beneficial cultures)
• Titratable acidity: ≥0.20% w/v (active fermentation marker)
• Residual reducing sugars: ≤5.0% w/v (fermentation completion indicator)
• TAMC: ≤1000 CFU/mL (elevated limit due to presence of cultural yeasts)
• TYMC: ≤500 CFU/mL (yeasts are normal microflora of Arishta/Asava)
• Coliform bacteria: <1 CFU/mL (zero tolerance)
• Salmonella: Absent in 25 mL (zero tolerance)
⚠️ WHY STANDARD MICROBIOLOGICAL LIMITS DO NOT APPLY:
Arishta and Asava are LIVE BIOTECHNOLOGICAL PREPARATIONS.
• Produced by natural fermentation (self-generation of alcohol)
• Contain beneficial yeasts and lactic acid bacteria as part of therapeutic matrix
• Are NOT sterile and SHOULD NOT be sterile
• Applying Ph.Eur. limits for non-aqueous liquids (<10² CFU/mL) would result in
false rejection of ALL legitimate Arishta/Asava batches
This utility uses SPECIFIC limits for fermented preparations,
based on understanding of their biological nature.
Usage:
AyurFermentedPreparationAlcoholAndMicrobiomeChecker.exe → demo mode
AyurFermentedPreparationAlcoholAndMicrobiomeChecker.exe input.csv output.json → evaluate data
Input format:
BatchNumber,ProductName,PrepType,Ethanol%,pH,TitratableAcidity%,ReducingSugars%,TAMC,TYMC,Coliforms,Salmonella
Example (Salmonella: false=absent, true=detected):
AYUR-001,Ashwagandharishta,Arishta,8.5,3.8,0.45,2.5,150,20,0,false
Preparation types:
• Arishta — fermentation from herbal decoction + jaggery/sugar
• Asava — fermentation from fresh herbal juice + jaggery/sugar
— PARAMETERS AND THEIR SIGNIFICANCE:
• Self-Generated Ethanol (5–12%):
Natural fermentation product. Serves as preservative and extractant.
<5%: insufficient preservation → spoilage risk during storage.
>12%: technology violation (possible external alcohol addition).
• pH (3.0–4.5):
Acidic environment created by organic acids during fermentation.
Suppresses pathogen growth, supports beneficial microorganisms.
Out of range → microbial balance disruption.
• Titratable Acidity (≥0.20%):
Total organic acid content (acetic, lactic, succinic).
Low acidity → incomplete or arrested fermentation.
• Residual Reducing Sugars (≤5.0%):
High sugars → fermentation incomplete → risk of secondary fermentation
in bottle during storage (swelling, turbidity, taste change).
• TAMC/TYMC (≤1000/≤500 CFU/mL):
Elevated limits reflect presence of cultural microorganisms.
Exceedance indicates contamination by foreign microflora.
• Coliforms & Salmonella:
ABSOLUTE ABSENCE. Presence = fecal contamination or GMP violation.
💡 Usage tips:
1. Sample aseptically but WITHOUT sample sterilization
2. Determine ethanol by gas chromatography or distillation + pycnometry
3. Perform microbiological plating WITHOUT prior ethanol neutralization
(1:10 dilution sufficient to reduce alcohol below inhibitory concentration)
4. For Asava (fresh juice based), expect higher TYMC than for Arishta
5. Monitor ethanol and sugar trends: they should be inversely correlated
6. Upon coliform/Salmonella detection — full investigation of water, raw materials and hygiene
⚠️ Note: This utility implements a fundamentally different microbiology approach
unfamiliar to Western pharmaceuticals. Instead of binary "sterile/non-sterile" it
evaluates the STATE OF THE LIVING ECOSYSTEM of the preparation. Fermented Ayurvedic
medicines are closer to kombucha or kefir than to tablets. Control must assess
ecosystem health, not attempt to destroy it.
