Water Microbiology Quality Checker — Control of Water Microbiological Purity

ℹ️  Utility checks microbiological quality of Purified Water (PW) and Water for Injection (WFI) according to Ph. Eur. 2.6.12 and USP <1231>:
     • TAMC (Total Aerobic Microbial Count): ≤ 100 CFU/mL
     • TYMC (Total Yeast and Mold Count): ≤ 10 CFU/mL
     • Pseudomonas aeruginosa: Absent in 100 mL
     • Coliforms: Absent in 100 mL
     • Strict control of incubation conditions (TAMC: 30-35°C, 3-5 days; TYMC: 20-25°C, 5-7 days)

⚠️  CRITICAL: Detection of Pseudomonas aeruginosa or coliforms is unacceptable!
     TAMC exceedance > 100 CFU/mL indicates biofilm in water purification system or sampling violation.

— WHY IS THIS NEEDED?
Water is a main component of most dosage forms. Microbiological contamination of water can lead to product spoilage, reduced efficacy, and patient risk (especially for parenterals).
Regular bioburden monitoring is necessary for validation and maintenance of the water purification system.

⚠️  CRITICAL:
• TAMC ≤ 100 CFU/mL — standard limit for PW and WFI at point of use.
• TYMC ≤ 10 CFU/mL — control of fungal contamination.
• Sampling: Must be performed aseptically after disinfection of sampling point and flushing sufficient volume.
• Incubation: Violation of temperature regime or incubation time invalidates the test.
• Action Levels: Facility may have stricter internal limits (e.g., 50 CFU/mL) for early warning.

💡 Usage tips:
1. Filtration: For water with low bioburden, membrane filtration method (filtering 100 mL or more) is recommended.
2. Media: Use R2A agar for TAMC (more sensitive to water bacteria than TSA) and SDA or DG18 for TYMC.
3. Neutralization: If water contains residual disinfectants (chlorine, ozone), use neutralizers in media or during filter rinsing.
4. Trends: Analyze data dynamically. A sharp spike in bioburden even within limits may indicate a problem.
5. Biofilm: Main source of contamination is biofilm in pipes. Regular system sanitization is mandatory.

⚠️ Note: Water microbiology is an indicator of the entire water preparation system status. A positive result requires immediate investigation and possibly re-sanitization of the loop.

URS & FS — Water Microbiology

This document describes the user requirements and functional specification for WaterMicrobiologyQualityChecker. The utility is intended for QC laboratory use as a rule-based check of data prepared from an instrument, LIMS, ELN, MES or an approved input.csv.

OQ/PQ scenarios

  • OQ-001: a valid sample row shall be processed without schema error.
  • OQ-002: a missing mandatory column shall produce a schema error.
  • OQ-003: a non-numeric value in a numeric field shall produce a conversion error.
  • OQ-004: a critical parameter outside the limit shall produce FAIL or a critical finding.
  • PQ-001: user real batches shall be checked with retention of input.csv, output.json, utility version and checksum.
