Unlike synthetic injections, herbal extracts contain saponins, polyphenols, proteins and polysaccharides that:
1. Cause false-negative LAL test results (gel-clot inhibition)
2. May possess pyrogenic activity independent of endotoxins
3. Create nutrient medium for microorganisms when sterilization is compromised
Without recovery validation, BET result in TCM is UNRELIABLE!

— WHY IS THIS NEEDED?
Parenteral administration bypasses all body protective barriers. For TCM this creates unique challenges:
• Complex multi-component composition cannot be fully characterized chemically
• Traditional sterilization methods (autoclaving) may destroy active components → manufacturers use filtration which is less reliable
• Herbal raw materials carry high initial bioburden
• Historically, TCM injections have caused serious adverse events (anaphylaxis, pyrogenic reactions)
Strict combined control of endotoxins + sterility + matrix validation is MANDATORY safety condition.
⚠️ CRITICAL:
• Endotoxin Limit = 5.0 EU/kg ÷ Max Dose (mL/kg) — individual calculation for each product
• BET Recovery 50–200% — WITHOUT this confirmation test is considered invalid for TCM
• Sterility: NO GROWTH in both media after 14 days
• Positive Control MUST pass — otherwise test is not sensitive to endotoxins in given matrix
• Negative Control MUST be clean — otherwise reagent/laboratory contamination
• Any sterility violation = AUTOMATIC BATCH REJECTION

💡 Usage tips:
1. For TCM injections ALWAYS perform preliminary BET validation with multiple dilutions
2. If Recovery <50%, use dilution, buffer neutralization or recombinant Factor C (rFC)
3. Sterility should be tested by membrane filtration method (to remove inhibitors)
4. Sterility incubation strictly 14 days at 30-35°C (TSB) and 20-25°C (FTM)
5. Upon any growth in sterility — microorganism identification is mandatory for CAPA
6. Track endotoxin trends: increasing values may indicate filter degradation or water system issues
⚠️ Note: This utility combines two critical safety tests that for TCM injections CANNOT be considered in isolation. High endotoxins with sterility indicate water/filtration problem. Growth with low endotoxins indicates aseptic technique breach. Low recovery with "normal" endotoxins indicates FALSE-NEGATIVE result. Only comprehensive assessment gives true safety picture of parenteral TCM product. This approach exceeds Ph.Eur. requirements where BET and sterility are often evaluated separately without consideration of herbal matrix specificity.

This document defines user requirements (URS) and functional specification (FS) for TcmEndotoxinAndSterilityQualityChecker. The utility is intended to evaluate input.csv data deterministically, generate structured output.json output and support traceable QA/QC review.
This document is a project-level URS/FS baseline for CSV/CSA, IQ/OQ/PQ and further validation under an approved laboratory procedure.
