SPR Binding Affinity Quality Checker — Control of Binding Affinity (SPR)

ℹ️  Utility assesses the kinetics of interaction of biological products (e.g., monoclonal antibodies) with their target using Surface Plasmon Resonance (SPR):
     • KD (Equilibrium Dissociation Constant) ≤ 1.0 nM (1.0E-9 M) — measure of binding strength
     • ka (Association Rate Constant) ≥ 1.0E5 1/Ms — rate of complex formation
     • kd (Dissociation Rate Constant) ≤ 1.0E-4 1/s — stability of the complex
     • Rmax (Maximum Response) 10–100 RU — control of activity and ligand density
     • Chi² (Chi-Squared) < 10% of Rmax — quality of fit to 1:1 Langmuir model
     • U-Value < 2.5 — statistical reliability of calculated parameters

⚠️  CRITICAL: High U-Value (>2.5) means parameters are determined with large error!
     Poor Chi² indicates that the model does not describe the data (possible aggregation or non-specific binding).

— WHY IS THIS NEEDED?
SPR (Surface Plasmon Resonance) is the gold standard for characterizing biosimilars and innovative biologics.
It allows determining not only binding strength (affinity) but also the kinetics of the process (how fast the drug binds and how long it stays on the target).
This is critically important for confirming efficacy and Mechanism of Action (MoA).

⚠️  CRITICAL:
• KD < 1 nM — typical requirement for high-affinity therapeutic antibodies.
• Rmax must be in optimal range: too low = poor sensitivity, too high = mass transport effects.
• Chi² and U-Value are Data Quality Metrics. If they are poor, KD/ka/kd values are unreliable, even if within limits.
• 1:1 Langmuir model assumes simple single-site binding. Deviations may indicate avidity or heterogeneity.

Critical parameters:
• KD: ≤ 1.0E-9 M
• ka: ≥ 1.0E5 1/Ms
• kd: ≤ 1.0E-4 1/s
• Rmax: 10.0 – 100.0 RU
• Chi²: < 10.0 (or <10% of Rmax)
• U-Value: < 2.5

💡 Usage tips:
1. Immobilization: Use ligand immobilization level ensuring Rmax in 10-100 RU range to avoid mass transport effects.
2. Reference: Always subtract reference cell (blank) signal to account for non-specific binding and baseline drift.
3. Model: If data is not described by 1:1 model (high Chi²), consider Bivalent Analyte or Heterogeneous Ligand models.
4. Concentrations: Use a series of analyte dilutions (usually 5-8 concentrations) around the expected KD.
5. Regeneration: Ensure chip surface is fully regenerated between cycles without loss of ligand activity.

⚠️ Note: SPR measures interaction in real-time without labeling (label-free). This provides the most physiologically relevant kinetic data compared to endpoint methods (ELISA).

URS & FS — SPR Binding Affinity

This document describes the user requirements and functional specification for SPRBindingAffinityQualityChecker. The utility is intended for QC laboratory use as a rule-based check of data prepared from an instrument, LIMS, ELN, MES or an approved input.csv.

OQ/PQ scenarios

  • OQ-001: a valid sample row shall be processed without schema error.
  • OQ-002: a missing mandatory column shall produce a schema error.
  • OQ-003: a non-numeric value in a numeric field shall produce a conversion error.
  • OQ-004: a critical parameter outside the limit shall produce FAIL or a critical finding.
  • PQ-001: user real batches shall be checked with retention of input.csv, output.json, utility version and checksum.
