Semisolid IVRT Quality Checker — In Vitro Release Testing for Semisolid Dosage Forms

ℹ️  Utility assesses the release kinetics of drug substances from ointments, creams, and gels according to USP <1724>:
     • Release Rate (must match reference product, typically 90-110%)
     • Method Precision (RSD ≤ 10% for a series of 6 cells)
     • Linearity of Higuchi plot (R² ≥ 0.98)
     • Lag Time (≤ 30 min)
     • Membrane Integrity (Critical System Suitability Parameter)

⚠️  CRITICAL: Membrane integrity failure completely invalidates test results!
     Non-linearity of the release plot indicates instability of the product structure or diffusion issues.

— WHY IS THIS NEEDED?
In Vitro Release Testing (IVRT) is a key quality control method for semisolid dosage forms (ointments, creams, gels).
It demonstrates bioequivalence of the product and stability of its manufacturing process.
According to FDA and USP guidelines, IVRT is used to confirm that changes in composition or process do not affect the availability of the active ingredient.

⚠️  CRITICAL:
• Release Rate — main criterion for comparison with reference product (Q1/Q2/Q3 sameness).
• RSD ≤ 10% — ensures reproducibility of the method between Franz cell units.
• Linearity (R² ≥ 0.98) — confirms that the process is diffusion-controlled (Higuchi kinetics).
• Membrane Integrity — if the membrane is damaged, data is invalid.
• Sink Conditions — 20-80% of the drug should be released for correct kinetic calculation.

💡 Usage tips:
1. Apparatus: Use vertical or horizontal Franz cells with controlled temperature (32±1°C for skin).
2. Membrane: Use synthetic membranes (e.g., PVDF or cellulose) inert to the product.
3. Degassing: Receptor solution must be degassed to prevent bubble formation under the membrane.
4. Sampling: Automated sampling is preferred to minimize errors and maintain volume.
5. Statistics: For new drug registration, comparison with a reference sample is required (f2 factor or confidence intervals).

⚠️ Note: IVRT does not measure skin penetration (which is done by IVPT), but it correlates with it and is the standard for quality control of semisolid manufacturing.

URS & FS — Semisolid IVRT

This document describes the user requirements and functional specification for SemisolidIVRTQualityChecker. The utility is intended for QC laboratory use as a rule-based check of data prepared from an instrument, LIMS, ELN, MES or an approved input.csv.

OQ/PQ scenarios

  • OQ-001: a valid sample row shall be processed without schema error.
  • OQ-002: a missing mandatory column shall produce a schema error.
  • OQ-003: a non-numeric value in a numeric field shall produce a conversion error.
  • OQ-004: a critical parameter outside the limit shall produce FAIL or a critical finding.
  • PQ-001: user real batches shall be checked with retention of input.csv, output.json, utility version and checksum.
