— WHY IS THIS NEEDED?
Methylphenidate — psychostimulant for ADHD and narcolepsy treatment:
• Often released as Extended Release (ER/XR) tablets to prevent concentration peaks and abuse
• Rapid release can lead to severe side effects (hypertension, tachycardia, psychosis)
• Synthesis may involve toluene (ICH Q3C Class 2)
• Specific impurities include phenylpiperidine derivatives
• Enantiomeric purity control important if dexmethylphenidate is used
⚠️ CRITICAL:
• Assay: 98–102%
• Melting Point: 58–62°C (for hydrochloride)
• Total Impurities: ≤ 0.5%
• Toluene: ≤ 890 ppm (strict limit)
• Dissolution (for ER): Strictly regulated ranges at 1h, 4h, 8h (e.g., 20-40%, 50-70%, >80%)
• Heavy Metals: ≤ 10 ppm
• Microbiology: standard limits
⚠️ Note: Utility adapted for dissolution profile check. If dissolution data absent (0), check skipped (for substance). For ER finished forms, deviation from dissolution profile is critical safety defect. Too rapid release dangerous due to overdose risk, too slow — lack of effect. Toluene control important due to neurotoxicity.

URS & FS — user requirements and functional specification

This document defines user requirements and functional specification for the quality-control utility. It supports deployment discussion, IQ/OQ preparation and QC workflow integration.

1. Scope

The Methylphenidate Quality Checker utility is used for Methylphenidate. The actual input.csv is the source of truth for the input structure; control criteria are defined by the executable and the domain description.

The utility does not use machine learning; decisions are produced by deterministic rules.

Limits stated in the description must be verified against the current approved specification, pharmacopoeial monograph and registration dossier before production use.

10. QA/QC, CSV and change control

  • Before production use, the client records executable version, checksum, specification/monograph version, test CSV, expected JSON and IQ/OQ results.
  • Column names must not be changed without updating the validator and test scenarios.
  • The source CSV, output.json and utility version should be stored together as an evidence package.
  • Any change in control rules must go through change control and repeated OQ scenario verification.
