Utility description: InfraLUM ID Fluconazole

InfraLUM ID Fluconazole — Fluconazole Identification by IR Spectroscopy

ℹ️ Utility checks IR spectrum compliance with reference (Ph. Eur. / Russian Pharmacopoeia):
    • Correlation coefficient with reference spectrum: ≥0.995
    • Peak 3150 cm⁻¹ (aromatic C-H stretching): ±15 cm⁻¹
    • Peak 1620 cm⁻¹ (triazole ring C=C): ±10 cm⁻¹ — CORE MARKER
    • Peak 1510 cm⁻¹ (fluorophenyl ring C=C): ±10 cm⁻¹
    • Peak 1270 cm⁻¹ (C-F stretching): ±10 cm⁻¹ — FLUORINE MARKER
    • Peak 1140 cm⁻¹ (triazole C-N): ±10 cm⁻¹
    • Baseline noise: ≤0.005 Abs

⚠️ IMPORTANT: Fluconazole is a systemic antifungal agent from the triazole class.
Structure contains two 1,2,4-triazole rings connected via a central carbon
to a 2,4-difluorophenyl group and a tertiary alcohol function.
Presence of two triazole rings gives intense C=N/C=C peaks in the 1600–1500 cm⁻¹ region.
Fluorine atom on the phenyl ring is a key structural element providing high lipophilicity
    and blood-brain barrier penetration.

Usage:
InfraLUM_ID_Fluconazole.exe → demo mode (console output)
  InfraLUM_ID_Fluconazole.exe input.csv output.json → evaluate your data

Input format:
BatchNumber,CorrelationCoefficient,Peak3150_cm1,Peak1620_cm1,Peak1510_cm1,Peak1270_cm1,Peak1140_cm1,BaselineNoiseAbs,PreparationMethod

Example (PreparationMethod: KBr / ATR / Nujol):
  FLU-ID-2026-041,0.9991,3148.0,1618.0,1512.0,1268.0,1142.0,0.0017,KBr

— WHY IS THIS NEEDED?
Fluconazole is one of the most prescribed antifungal drugs.
• Mechanism of action: inhibition of ergosterol synthesis in fungal cell walls.
• IR spectroscopy allows rapid confirmation of substance identity.
• Reference spectrum established in Ph. Eur. Fluconazole and Russian Pharmacopoeia FS.2.4.00189.18.
• InfraLUM FT-08/FT-10 instruments are widely used in Russian QC laboratories.

⚠️ CRITICAL:
• Correlation ≥0.995 — strict matching criterion with reference.
• Peak 1620 cm⁻¹ (triazole C=C) — primary heterocyclic core marker.
Absence indicates triazole ring destruction or substitution.
• Peak 1270 cm⁻¹ (C-F) — specific marker for fluorinated azoles.
Absence of C-F peak may indicate ketoconazole or clotrimazole (non-fluorinated).
• Peak 1140 cm⁻¹ (C-N) — confirms nitrogen-carbon bond in triazole.
• Peak pair 1620/1510 cm⁻¹ reflects conjugated triazole-phenyl system.
• Noise >0.005 Abs renders spectrum unsuitable for identification.

Key features:
• Uses NAMED TUPLE (record struct EvalResult) —
  eliminates positional field access errors.
• Specification based on Ph. Eur. Fluconazole and Russian Pharmacopoeia FS.2.4.00189.18.
• Control of five characteristic peaks + correlation + spectrum quality.
• Built-in differentiation from non-fluorinated azoles via C-F peak.
• Full LabWare LIMS compatibility.

Critical parameters:
• Correlation Coefficient: ≥0.995
• Peak 3150 cm⁻¹ (arom. C-H): 3150 ± 15 cm⁻¹
• Peak 1620 cm⁻¹ (triazole C=C): 1620 ± 10 cm⁻¹
• Peak 1510 cm⁻¹ (fluorophenyl C=C): 1510 ± 10 cm⁻¹
• Peak 1270 cm⁻¹ (C-F stretch): 1270 ± 10 cm⁻¹
• Peak 1140 cm⁻¹ (triazole C-N): 1140 ± 10 cm⁻¹
• Baseline Noise: ≤0.005 Abs

💡 Usage tips:
1. KBr tablets must be dry — pre-dry at 120°C before use.
2. Sample concentration in KBr: 0.5–1.0% (w/w).
3. Record spectrum in 4000–400 cm⁻¹ range, 4 cm⁻¹ resolution, 32 scans.
4. Record background spectrum of pure KBr before each measurement.
5. For ATR method, ensure tight crystal-sample contact.
6. Reference spectrum should be obtained on the same instrument under identical conditions.
7. Fluconazole is thermally stable but hygroscopic — store in a dry place.

⚠️ Note: Fluconazole exists as a racemate. IR spectroscopy does NOT distinguish
enantiomers. However, the molecule is symmetric relative to the central carbon,
simplifying spectrum interpretation. The tertiary alcohol group (-OH) gives
a broad band in the 3200–3400 cm⁻¹ region, often overlapping with atmospheric
moisture or trace water in KBr. During analysis, pay attention to the shape
of this band: narrow peak indicates free OH, broad indicates hydrogen bonding.
Difluoro substitution on the phenyl ring also appears in the 800–900 cm⁻¹ region
(C-H out-of-plane), serving as an additional confirmatory marker.
