Utility description: Ibuprofen Related Substances Checker

Ibuprofen Related Substances Checker — Ibuprofen (Related Substances Control)

ℹ️ Utility checks impurity profile per Ph. Eur. / Russian Pharmacopoeia:
    • Impurity A (4-isobutylbenzoic acid): ≤0.1%
    • Impurity B (4-isobutylacetophenone): ≤0.1%
    • Impurity C (positional isomer): ≤0.1%
    • Impurity D (1-(4-isobutylphenyl)ethanol): ≤0.1%
    • Any other single impurity: ≤0.1%
    • Total impurities: ≤0.3%
    • Assay: 99.0–101.0%
    • R-enantiomer (inactive): ≤3.0%

⚠️ IMPORTANT: Ibuprofen is a chiral molecule (S/R racemate).
    Only S-enantiomer has anti-inflammatory activity.
R-enantiomer is inactive and controlled as stereochemical impurity.
Impurities A and B are key synthesis quality markers (oxidation, incomplete conversion).

Usage:
IbuprofenRelatedSubstancesChecker.exe → demo mode (console output)
  IbuprofenRelatedSubstancesChecker.exe input.csv output.json → evaluate your data

Input format:
BatchNumber,ImpurityAPercent,ImpurityBPercent,ImpurityCPercent,ImpurityDPercent,AnyOtherSingleImpurityPercent,TotalImpuritiesPercent,AssayPercent,EnantiomericPurityPercent

Example:
  IBU-RS-2026-044,0.04,0.03,0.02,0.01,0.05,0.18,99.7,1.5

— WHY IS THIS NEEDED?
Ibuprofen is an NSAID, propionic acid derivative.
• Synthesis includes Friedel-Crafts acylation and reduction steps,
creating risk of residual ketones and alcohols (impurities B, D).
• 4-Isobutylbenzoic acid (impurity A) is an oxidation product during storage.
• Positional isomers (impurity C) form when regioselectivity is violated.
• Chiral control is mandatory: pharmacopoeial ibuprofen is a racemate,
but inactive R-enantiomer content is limited.

⚠️ CRITICAL:
• Each known impurity ≤0.1% — ICH Q3A qualification threshold.
• Total impurities ≤0.3% — general API purity limit.
• R-enantiomer ≤3.0% — stereochemical purity (Ph. Eur.).
• Assay 99.0–101.0% — dosage assurance.

Key features:
• Uses NAMED TUPLE (record struct EvalResult) —
  eliminates positional field access errors.
• Specification based on Ph. Eur. Ibuprofen and Russian Pharmacopoeia FS.2.4.00178.18.
• Control of four specific impurities + unknowns + enantiomer.
• Full LabWare LIMS compatibility.

Critical parameters:
• Impurity A (4-isobutylbenzoic acid): ≤0.1%
• Impurity B (4-isobutylacetophenone): ≤0.1%
• Impurity C (positional isomer): ≤0.1%
• Impurity D (1-(4-isobutylphenyl)ethanol): ≤0.1%
• Any Other Single Impurity: ≤0.1%
• Total Impurities: ≤0.3%
• Assay: 99.0–101.0%
• R-Enantiomer: ≤3.0%

💡 Usage tips:
1. Impurities determined by HPLC with UV detection (214 nm).
2. Enantiomeric purity determined on chiral column (normal phase).
3. Impurity A may increase during storage — monitor conditions (temperature, light).
4. Use Ph. Eur. CRS reference standards for quantification.
5. Dissolve sample immediately before analysis (solution unstable >24 h).

⚠️ Note: Ibuprofen is prone to dimer and ester formation at elevated temperatures.
If unknown peaks >0.1% are detected, LC-MS identification and qualification
    per ICH Q3A(R2) are required.
