— WHY IS THIS NEEDED?
Entecavir — guanosine nucleoside analog for chronic hepatitis B treatment:
• Chiral molecule: activity depends on enantiomeric purity (specific rotation control)
• Usually exists as monohydrate, so loss on drying must be in strict range (4-6%)
• Poorly soluble in water, so dissolution control critical for bioavailability and virological response
• Synthesis may involve DMF (ICH Q3C Class 2)
• Specific impurities include dehydration products and epimers
• Drug taken chronically (often lifelong), so heavy metal requirements are stricter than usual

• Assay: 98–102%
• Specific Rotation: -55° to -45° (for active enantiomer)
• Melting Point: 248–252°C
• Loss on Drying: 4.0–6.0% (confirmation of monohydrate status)
• Total Impurities: ≤ 0.5%
• DMF: ≤ 880 ppm (strict limit)
• Heavy Metals: ≤ 10 ppm
• Dissolution: ≥ 80% in 30 min, ≥ 85% in 60 min (to ensure stable viral suppression)
• Microbiology: standard limits
⚠️ Note: Utility adapted for dissolution profile and hydration status check. If dissolution data absent (0), check skipped (for substance). For finished forms, deviation from dissolution profile is critical defect as it affects drug blood levels and risk of viral resistance. Specific rotation control important for confirming chiral purity. LOD check critical: too low value indicates loss of crystallization water (polymorph change), too high — presence of free moisture.

URS & FS — user requirements and functional specification

This document defines user requirements and functional specification for the quality-control utility. It supports deployment discussion, IQ/OQ preparation and QC workflow integration.

1. Scope

The Entecavir Quality Checker utility is used for Entecavir. The actual input.csv is the source of truth for the input structure; control criteria are defined by the executable and the domain description.

The utility does not use machine learning; decisions are produced by deterministic rules.

4. Domain limits and critical parameters

10. QA/QC, CSV and change control

  • Before production use, the client records executable version, checksum, specification/monograph version, test CSV, expected JSON and IQ/OQ results.
  • Column names must not be changed without updating the validator and test scenarios.
  • The source CSV, output.json and utility version should be stored together as an evidence package.
  • Any change in control rules must go through change control and repeated OQ scenario verification.
