ICH Q3D guideline and pharmacopoeias (USP <232>, Ph. Eur. 5.20) require assessment and control of elemental impurities. • Impurities may originate from catalysts, reagents, water, equipment, or packaging. • Element toxicity is assessed via Permitted Daily Exposure (PDE). • ICP-MS (Inductively Coupled Plasma Mass Spectrometry) methods provide necessary sensitivity (ppb/ppm level). • Automatic limit calculation based on daily dose allows flexible application for different drugs. ⚠️ CRITICAL: • Route of administration (oral, parenteral, inhalation) affects PDE values. This utility uses conservative oral defaults. • Sample preparation (microwave digestion) is critical for complete metal dissolution. • Spectral and matrix interferences must be controlled during analysis. • Use of internal standards (ISTD) is mandatory for instrument drift correction. Key features: • Calculation of individual limits based on MDD. • Check of Class 1 and 2A elements. • Support for ICP-MS and ICP-OES data. Always specify the accurate Maximum Daily Dose (MDD) of the product. 2. For parenteral products, use corresponding, stricter PDE values. 3. Regularly calibrate the instrument using multi-element standards. 4. Verify method Limits of Detection (LOD) and Quantitation (LOQ). 5. Maintain a logbook for plasma source and mass filter maintenance. ⚠️ Note: Some elements (e.g., arsenic and mercury) exist in different chemical forms (speciation) which may have different toxicities. ICP-MS determines total element content.

This document describes the user requirements and functional specification for ElementalImpuritiesICPQualityChecker. The utility is intended for QC laboratory use as a rule-based check of data prepared from an instrument, LIMS, ELN, MES or an approved input.csv.

OQ/PQ scenarios
