Circular Dichroism Biologic Quality Checker — Control of Biologic Structure (CD)

ℹ️  Utility checks critical parameters of protein molecule conformation:
   • Secondary structure (ellipticity at 222 nm): within ±10% of reference
   • Tertiary structure (ellipticity at 280-295 nm): within ±10% of reference
   • Thermal stability (Tm): decrease no more than 2°C
   • Spectral correlation: ≥0.99

⚠️  CRITICAL: Changes in CD spectrum indicate denaturation or misfolding!
   Decrease in Tm indicates loss of product stability during storage.

— WHY IS THIS NEEDED?
For biological products (monoclonal antibodies, recombinant proteins, vaccines), structure determines function.
• Circular Dichroism (CD) in far-UV range (190-250 nm) is sensitive to secondary structure (alpha-helices, beta-sheets).
• Near-UV range (250-320 nm) reflects the environment of aromatic amino acids and disulfide bonds (tertiary structure).
• Thermal denaturation with CD monitoring allows determination of melting temperature (Tm) — a key stability parameter.
• USP <1053> and ICH Q6B guidelines recommend using spectroscopic methods for higher-order structure characterization.

⚠️  CRITICAL:
• Even small changes in ellipticity may indicate loss of activity.
• Decrease in Tm can lead to product aggregation during transport or storage.
• High correlation with reference spectrum confirms conformational identity.
• Method requires high buffer purity and absence of absorbing impurities.

• Support for circular dichroism spectrometer data (e.g., JASCO J-1500).

Critical parameters:
• Secondary Structure Ellipticity: +/- 10% of Ref
• Tertiary Structure Ellipticity: +/- 10% of Ref
• Tm: >= Ref Tm - 2°C
• Spectral Correlation: >= 0.99

💡 Usage tips:
1. Use quartz cuvettes with short optical path length (0.1–1 mm) for far-UV.
2. Carefully subtract buffer spectrum (baseline subtraction).
3. For Tm determination, perform heating with controlled rate (e.g., 1°C/min).
4. Protein concentration must be accurately known for molar ellipticity calculation.
5. Compare spectra with reference standard of the specific molecule measured under identical conditions.

⚠️ Note: CD is an integral method providing information about the average state of all molecules in solution. It does not resolve individual conformers like NMR, but is ideal for rapid quality control and batch comparison (fingerprinting).

URS & FS — Circular Dichroism Biologic

This document describes the user requirements and functional specification for CircularDichroismBiologicQualityChecker. The utility is intended for QC laboratory use as a rule-based check of data prepared from an instrument, LIMS, ELN, MES or an approved input.csv.

OQ/PQ scenarios

  • OQ-001: a valid sample row shall be processed without schema error.
  • OQ-002: a missing mandatory column shall produce a schema error.
  • OQ-003: a non-numeric value in a numeric field shall produce a conversion error.
  • OQ-004: a critical parameter outside the limit shall produce FAIL or a critical finding.
  • PQ-001: user real batches shall be checked with retention of input.csv, output.json, utility version and checksum.
