Capillary ID Aprotinin — Aprotinin Identification (CE)

ℹ️  Utility checks capillary electrophoresis parameters for aprotinin:

— WHY IS THIS NEEDED?
Aprotinin is a polypeptide drug used as a hemostatic agent (kallikrein and plasmin inhibitor).
Capillary Electrophoresis (CE) is highly effective for separating charged protein molecules and their degradation forms.
• Identification is performed by migration time relative to a standard sample.
• Anhydroaprotinin is formed by dehydration of the molecule and is a key impurity.
• Control of separation efficiency (theoretical plates) and peak symmetry is critical for CE method validation.

⚠️  CRITICAL:
• Migration time must strictly match the standard (window ±0.5 min).
• Main substance purity ≥98.0%.
• Anhydroaprotinin content ≤1.0%.
• System must ensure high efficiency (>10000 plates) for clear separation of isoforms.

💡 Usage tips:
1. Use coated capillaries to minimize protein adsorption on walls.
2. Buffer solution should contain additives (e.g., urea or surfactants) to prevent aggregation.
3. Condition the capillary before each run.
4. Monitor thermostat temperature as buffer viscosity affects migration time.

⚠️ Note: Unlike small molecules, proteins in CE can yield broad peaks due to multiple charges and conformational states. Strict tailing control is mandatory.

URS & FS — User Requirements and Functional Specification

This document defines the controlled interface, validation behaviour and expected output of Capillary_ID_Aprotinin. The document is intended to reside inside the utility folder together with input.csv and the description file. Expected executable: Capillary_ID_Aprotinin.exe.

Purpose: the utility performs deterministic verification of QC input data against approved limits, generates machine-readable output.json, and supports QC/QA review, LIMS/ELN/MES integration and CSV/CSA preparation.

QA/QC, CSV/CSA and change control

  • Do not rename columns without updating the validator, test set and documentation.

  • Before production use, perform IQ/OQ/PQ or equivalent risk-based CSV/CSA verification.
  • Critical limits shall be approved by the process owner and verified against the registration dossier, pharmacopoeia and local SOPs.
