AutomatedLiquidHandlingQCQualityChecker
Automated Liquid Handling QC
Automated Liquid Handling QC Quality Checker — Quality Control for Automated Liquid Handling Systems ℹ️ Utility checks critical parameters of automated liquid handlers (Hamilton, Tecan, Eppendorf, etc.): • Accuracy (Systematic Error) — deviation of mean volume from target • Precision (CV / Random Error) — reproducibility of dispensing • Compliance with ISO 8655 and USP <1058> (instrument qualification) ⚠️ CRITICAL: Dispensing errors >3% (accuracy) or CV >1% (precision) can lead to incorrect results in HPLC, ICP-MS, ELISA, and other assays! Usage: AutomatedLiquidHandlingQCQualityChecker.exe → demo mode (console output) AutomatedLiquidHandlingQCQualityChecker.exe input.csv output.json → evaluate your data Input format: BatchNumber,TargetVolumeUL,MeanMeasuredVolumeUL,StandardDeviationUL,NumberOfReplicates,WaterDensityMGPerUL Example: CAL-HAMILTON-2026-001,100.0,99.8,0.4,10,0.9982 Supported equipment types: • Automated Workstations (Hamilton STAR, Tecan Fluent, Beckman Biomek) • Liquid Handlers (Eppendorf epMotion, Gilson Prep) • Calibration Events (Regular gravimetric calibration) — WHY IS THIS NEEDED? Automated dispensing is the foundation of reproducibility in modern pharma labs: • Sample preparation for elemental analysis (ICP-MS) requires high dilution accuracy • Microplate setup for ELISA/biological tests requires low variability (CV) • USP <1058> requires regular qualification (PQ) of all measurement systems, including pipettes and handlers ⚠️ CRITICAL: • Accuracy: Deviation of mean volume from target. Limit typically ±3% (volume-dependent). • Precision (CV): Coefficient of variation. Limit typically <1% for volumes >50 µL. • Gravimetric method: Primary verification method. Uses water density (Z-factor) to convert mass to volume. • Temperature and humidity: Must be controlled during calibration (ISO 8655). Key features: • Automatic calculation of % Error (Accuracy) and Coefficient of Variation (CV) • Comparison with configurable pharmacopoeial/internal limits • Support for gravimetric data (mass -> volume via density) • LIMS-compatible report generation (LabWare) for qualification status (Pass/Fail) Critical parameters: • Accuracy: Within ±3.0% (example for 100 µL) • Precision (CV): ≤ 1.0% (example for 100 µL) • Water Density: Temperature-corrected (e.g., 0.9982 mg/µL at 20°C) 💡 Usage tips: Perform gravimetric calibration at least quarterly or after maintenance. Use analytical balances with appropriate weighing range (minimum weight). For small volumes (<10 µL), use evaporation traps and specialized techniques. If CV is high — check tips, seals, and air bubbles. If Accuracy is low — perform software calibration (calibration factor) of the handler. ⚠️ Note: This utility does not control the robot directly but analyzes gravimetric test data loaded by the operator or exported from balance software. It is a key element of the laboratory's Quality Assurance (QA) system.

URS & FS — Automated Liquid Handling QC
This document describes the user requirements and functional specification for AutomatedLiquidHandlingQCQualityChecker. The utility is intended for QC laboratory use as a rule-based check of data prepared from an instrument, LIMS, ELN, MES or an approved input.csv.
URS — User Requirements Specification
| ID | Requirement | Criticality | Acceptance criterion |
|---|---|---|---|
| URS-001 | The utility shall accept input.csv with approved columns: BatchNumber, TargetVolumeUL, MeanMeasuredVolumeUL, StandardDeviationUL, NumberOfReplicates, WaterDensityMGPerUL. | High | The CSV file is processed without manual header editing. |
| URS-002 | The utility shall perform deterministic evaluation for “Automated Liquid Handling QC” without machine learning. | High | The same input data produce the same JSON result. |
| URS-003 | The utility shall validate mandatory fields, numeric formats, flags, ranges and domain plausibility. | High | Schema and conversion errors are explicitly reported. |
| URS-004 | The utility shall generate output.json with PASS / WARNING / FAIL statuses, source values, warnings and failures. | High | The JSON result is suitable for review, deviation investigation and integration. |
| URS-005 | The documentation shall support IQ/OQ/PQ or CSV/CSA verification. | Medium | URS/FS, CSV/JSON contract and test scenarios are supplied with the utility. |
input.csv contract
