AAS Glass Leachables Test — Evaluation of Leachables from Glass Containers

ℹ️  Utility checks critical quality parameters of glass containers (USP <660> / Ph. Eur. 3.2.1):
   • Lead (Pb): ≤5.0 ppm (in extract)
   • Cadmium (Cd): ≤0.5 ppm (in extract)
   • Arsenic (As): ≤0.5 ppm (in extract)
   • Alkalinity (Hydrolytic Resistance): ≤10.0 mL 0.01M HCl (for Type I Glass)

⚠️  CRITICAL: High alkalinity (>10.0 mL) → glass does not meet Type I criteria!
   Risk of product pH shift and precipitate formation (glass flakes).

— WHY IS THIS NEEDED?
Glass containers (vials, ampoules, cartridges) contact the drug product.
• Glass can leach metal ions and alkali, especially during sterilization and storage.
• USP <660> and Ph. Eur. 3.2.1 classify glass into Types I, II, III, NP based on hydrolytic resistance.
• Type I (borosilicate) is the most inert, used for parenteral products.
• Atomic Absorption Spectrometry (AAS) on Lumex MGA-1000 allows precise determination of trace Pb, Cd, As in water extract.
• Titration determines the amount of released alkali (hydrolysis of silicate network).

⚠️  CRITICAL:
• Alkalinity ≤10.0 mL 0.01M HCl — criterion for Type I Glass compliance (Powdered Glass Test).
• Lead, Cadmium, Arsenic are toxic elements; their leaching must be minimal.
• Glass degradation ("delamination") can lead to visible particles in the product.
• Control is necessary for each batch of primary packaging.

💡 Usage tips:
1. For Powdered Glass Test, use glass fraction 300-500 µm.
2. Perform extraction in autoclave at 121°C for 30 minutes.
3. For alkalinity determination, use precise titration with 0.01M HCl and indicator (methyl red).
4. For metal determination, use Graphite Furnace AAS (GFAAS) on MGA-1000 for high sensitivity.
5. If glass fails Powdered Glass Test, it cannot be classified as Type I.

⚠️ Note: Leaching depends on surface area contact. Powdered Glass Test is accelerated and stricter than Water Attack at 121°C test, which is applied to finished containers.

URS & FS — User Requirements and Functional Specification

This document describes the controlled interface, user requirements and functional behaviour of AAS_Glass_Leachables_Test. The utility is intended for automated verification of laboratory, pharmacopoeial, analytical or manufacturing QC parameters using input.csv and producing a structured output.json result.
